Carbohydrate Name
Inner core Lipopolysaccharide
Carbohydrate Class
Core oligosaccharide [LPS]
Source Microbe
Escherichia coli F653
Basic Structure
It consists of the uncommon sugars L-glycero-D mannoheptose (Hep) and 3-deoxy-D-manno-octulosonic acid (Kdo) along with Lipid A
Proposed functions
N/A
Antigenic Nature used to produce antibodies
Heat killed bacteria
Carrier Name
nil
Conjugation Method
nil
Antibodies
Mab WN1 222-5
Antibody type and class
IgG2a
Assay System
ELISA, immunoblotting, passive hemolysis assay
Cross-reactivity
WN1 222-5 binds to all E. coli clinical isolates tested so far (79 blood isolates, 80 urinary isolates, and 21 fecal isolates), Salmonella LPS, Shigella LPS and to some Citrobacter, Enterobacter, and KiebsieUla isolates
Proposed epitopes
The epitope consists of the inner core
Proposed Utility
In vitro, this Mab inhibits the Limulus amoebocyte lysate assay activity of S-form and R-form LPS in a dose-dependent manner and it neutralizes the LPS-induced release of clinically relevant monokines (interleukin 6 and tumor necrosis factor). In vivo, WN1 222-5 blocks endotoxin-induced pyrogenicity in rabbits and lethality in galactosamine-sensitized mice
