Carbohydrate Name

O-polysaccharide

Carbohydrate Class

O-polysaccharide [LPS]

Source Microbe

Actinobacillus pleuropneumoniae serotype 4 (field isolate F6)

Basic Structure

O chains of LPS of serotypes 4 and 7 were structurally similar, being composed of repeating tetrasaccharide units with identical trisaccharide residues forming a common backbone

Proposed functions

LPS is an important virulence factor. It molecule plays an important role in adherence of the bacterium to porcine respiratory tract cells and mucus

Antigenic Nature used to produce antibodies

formalin-fixed A. pleuropneumoniae serotype 4 cells

Carrier Name

nil

Conjugation Method

nil

Antibodies

Mab 3E12

Antibody type and class

IgM

Assay System

Whole cell ELISA and slide agglutination tests

Cross-reactivity

This Mab failed to recognize surface epitopes located on other bacterial, such as H. parasuis, P. haemolytica, P. multocida capsular types A and D, Haemophilus taxon minor group, and A. suis, K. pneumoniae and B. bronchiseptica. were reactive with 26 serotype 4 isolates,11 serotype 7 isolates, and 1 each of the serotype 1 and non-typeable isolates tested

Proposed epitopes

N/A

Proposed Utility

This MAb could be useful for the serological diagnostic of animals infected with serotypes 4 and 7 by inhibition ELISA or competition ELISA. Moreover, they could be useful as complementary reagents to polyclonal sera for serotyping of field isolates belonging to A. pleuropneumoniae serotype 4 or 7
